Testosterone promotes the commitment of mesenchymal pluripotent cells into myogenic lineage, the hypothesis that AR is expressed in mesenchymal cells in skeletal muscle precursors. AR expression was evaluated by immunohistochemical staining, confocal immunofluorescence and immunoelectron microscopy in sections of vastus

lateral healthy men before and after treatment with a supraphysiological dose of testosterone enanthate. Crops satellite cells in skeletal muscle of humans were also tested for AR expression. AR protein was expressed mainly in cells satellite, identified by their location on the outside and inside the sarcolemma basement membrane, and CD34 and c-MET staining. Myonuclei in many muscle fibers also showed AR immunostaining. Furthermore, CD34 + stem cells in the interstitium, fibroblasts, and mast cells expressed AR immunoreactivity. Immunoelectron microscopy revealed the aggregation of particles in nucleoli of satellite immune cells and myonuclei, testosterone treatment increased nucleolar AR density. AR mRNA and protein expression in satellite cell cultures was confirmed by RT-PCR, reverse transcription and real-time PCR, the sequence of the RT-PCR and product analysis by Western blot.
